How do I make a sprite?

I know, it's probably a dumb question, but I was curious if there was a set way to go about generating a sprite, or pixelated character set, ala Cloudsfactory? I'm looking to try my had at a character set from some of my favorite books, but I'm not quite sure how to go about it. What do you guys do to generate it? Do you use a software, or do you do it completely zoomed in to see the grid in software like MS Paint? Just curious!

I use KG Cross Stitch or PC Stitch. I've also done the zoomed in and draw it in Excel (you can resize the boxes to be square) as well as on graph paper back in the olden days

I use paint.net and have a grid displayed. I then use the pencil tool and color one square in at a time.

Yeah, under the words "Effects" there's a grid display button. You have to be zoomed in in order to see the grid.

In Photoshop you can do it too!
